C57BL / 6J 超排小鼠取卵时间对体外受精效果的影响

Effect of Collecting Oocyte on Vitrofertilization in C57BL / 6J Superovulated Mice

摘要: 目的 探讨雌性小鼠注射绒毛膜促性腺激素( HCG)后,取卵时间对体外受精率的影响。 方法 选用 3 ~ 4 周龄的野生型 C57BL / 6J 雌鼠,体质量 10 ~ 13 g,通过腹腔注射血清促性腺激素( PMSG) 和 HCG 联合使用[ 1] 做超排处理。 我们将超排后雌鼠取卵时间分为 6 个时间段,分别为 13、14、15、16、17、18 h 后取卵母细胞与新鲜精子做体外受精。 取 3 月龄雄性小鼠附睾里精子,在 HTF 液里获能 1 h 后,用于体外受精。 实验共分 3 组,每组 12 只雌鼠用于超排处理,共得到 2-细胞胚胎数分别为 258、199 和 243 枚;分别移植到当天见栓的假孕鼠输卵管内,得到出生仔鼠分别为 98 只、87 只、95 只;出生率分别为 37. 98%、43. 72% 和 39. 09%。 结果 总取卵数和 2-细胞发育率,超排后15 h 取卵发育受精率最高,这说明小鼠卵母细胞超排后 14 h 少数成熟,15 ~ 16 h 完全成熟,排卵 17 h 后则开始出现退化。 结论 3 组实验最高受精率比较:第 1 组 15 h 后取的卵母细胞团受精率最高为 79. 17%、第 2 组 15 h 后取的卵母细胞团受精率最高为 75. 68%、第 3 组 16 h 后取的卵母细胞团受精率最高为 76. 92%。

Abstract: Collecting Oocyte time on IVF rate after HCG injection. Method 3-4 week-old wild-type C57BL / 6J females with 10-13 g were treated with PMSG and HCG. We divided the egg retrieval time of female mice after superarrangement into six periods, after 13, 14, 15, 16, 17 and 18 h for in vitro fertilization. Sperm from the epididymis of 3-month-old male mice were recovered in HTF fluid for 1 h and then used for in vitro fertilization. The experiment was divided into 3 groups, and 12 females were used for overarrangement, and 258,199 and 243 embryos were obtained. In the experiment, 98,87 and 95 births were obtained; the birth rates were 37. 98%, 43. 72% and 39. 09% respectively. Result The total number of eggs and the development rate of 2 cells were the highest at 15 h after superovulation, which showed that the mouse oocytes matured after 14 h, fully mature at 15 to 16 h, and began to degenerate after 17 h of ovulation. Conclusion Comparison of the highest fertilization rate in the 3 groups: the oocyte mass fertilization rate after 15 h in the first group was 79. 17%, the oocyte mass fertilization rate in the second group after 15 h was 75. 68%, and the oocyte mass fertilization rate after
16 h in the third group was 76. 92%.
